New Inhaled Insulin Study
Bioavailabilty of the oral insulin was 1.8 times
that of insulin injected subcutaneously
BioSante Pharmaceuticals, Inc.announced that positive
study results of its BioAir(TM) calcium phosphate
nanoparticulate (CAP) delivery system for inhalable
insulin have been published in the scientific journal
of the American Association of Pharmaceutical Scientists
(AAPS), AAPS PharmSci.
"Our advanced CAP technology not only allows
certain medications to be inhaled rather than injected,
but may substantially reduce the size and frequency
of dosing required," said Stephen M. Simes,
president and chief executive officer of BioSante.
He noted that the innovative technology is also being
used to develop insulin that can be administered
orally rather than by injection.
The new study, conducted by scientists at the University
of North Carolina and BioSante, evaluated an insulin
formulation using CAP and polyethylene glycol (PEG)
particles in suspension for administration via inhalation.
The insulin-CAP-PEG particles were administered
to the lungs of fasted rats. The study demonstrated
that CAP-PEG particles significantly reduced the
elimination of insulin, increasing its systemic residence
time and duration of action. Consequently, the amount
of insulin that became available through the bloodstream
(bioavailability) using the BioAir insulin-CAP-PEG
formulation was 1.8 times that of insulin injected
subcutaneously.
"The reduced bioavailability observed in studies
with previous delivery systems for inhaled insulin
has been a major concern, and had been called insurmountable," said
Dr. Lucila Garcia-Contreras, research assistant professor
at the University of North Carolina, one of the researchers. "Results
of this study may support the potential use of insulin-CAP-PEG
particles to increase the effectiveness of drug delivery
through the lung. The CAP technology consists of
microscopic particles of a natural compound similar
to that found in teeth and bones. The patented formulation
is nontoxic and biodegradable, offering a multitude
of potential medical applications.
